When high temporal resolution is critical, so is the selection of a “continuous” system for indirect calorimetry measurements. Similar to the Promethion multiplexed systems, this high-definition continuous system for mice combines ultra-high resolution with the depth-of-field array of synchronously acquired measurements that include energy expenditure, RQ, VO2, VCO2 and activity. Superior cage design – created by scientists, for scientists – assures both accuracy and animal enrichment while avoiding “dead space” compromises. Unlike the multiplexed system, however, the continuous system measures all metabolic parameters from all cages simultaneously – each cage has its own dedicated gas analysis system with a record-breaking one second resolution, identical to the temporal resolution of the cage sensors – including position, intake, wheel running, body mass and more. This creates a richer and higher-resolution metabolic data stream, while retaining secure GLP compliant raw data storage and analytical flexibility. Flow rate is in compliance with IACUC air exchange standards.
Given the unrivaled sample speed of the Promethion multiplexed calorimetry and behavioral phenotyping system, the key to selecting a continuous system is the temporal resolution your application demands. The additional investment in a continuous system is best justified if your application requires tracking subtle and rapid metabolic changes, such as those accompanying specific activities or caused by administering specific drugs or stimuli. Such an insightful data stream can reveal unexpected correlations or expose assumptions hidden in the gaps of a multiplexed system.

The chart above shows the temporal resolution of a typical competitor’s multiplexed system in which several cages share a gas analysis chain (red line), compared to our new parallel, continuous system (black line). Promethion accurately tracks the energy expenditure accompanying even brief episodes of activity or inactivity, enabling quantification of rapid, subtle metabolic signals from multiple animals simultaneously with more accurate allocation of energy costs to specific behaviors than previously possible.
Promethion’s high resolution activity detection arrays capture actual activity and discard noise, giving an accurate indication of real activity (orange trace at the bottom of the graph). Body mass is shown as the black line at the top of the graph and energy expenditure (EE) is in blue. You can see that the EE drops to a low, relatively stable level when the mouse (in this case a 22 g C57BL/6J male) is inactive. The small oscillations visible in EE during inactivity are real, and are not detectable by lower-resolution systems. Then, with each bout of activity, you can see EE rising to a relatively stable level and falling back down to resting levels after the activity bout ceases. Due to Promethion’s high resolution, there is no blurring or smearing of the data.
